银杏叶提取物对大鼠肝硬化的治疗作用及其机制研究

       研究发现肝细胞凋亡参与了多种急慢性肝脏疾病的发生、发展过程,如病毒性肝炎、酒精性肝病、非酒精性脂肪肝、胆汁淤积性肝病及缺血一再灌注损伤等等,是肝细胞损伤的重要机制之一〔12-17〕。体外及体内试验发现二甲基亚硝胺、放线菌酮、硫代乙酰胺等常见的肝毒性药物,均可诱导肝细胞发生凋亡〔18〕。近年〔19〕等研究发现,给鼠喂饲四氯化碳产生的经典肝损伤动物模型,运用流式细胞仪、透射电镜、电泳及原位末端标记等技术均检测到这种动物模型肝损伤的肝脏存在细胞凋亡。研究还发现,病毒性肝炎肝损伤时,凋亡与坏死同时存在,或凋亡的发生早于坏死〔20〕;临床各期酒精性肝病患者活检标本均发现有肝细胞凋亡。因此,肝细胞凋亡在肝脏疾病发病机制中具有重要作用,抑制肝细胞的凋亡有助于延缓慢性肝病的发生和发展。本试验研究发现,肝硬化大鼠经口服给予EGb后,能降低肝细胞凋亡率,抑制肝细胞的凋亡,抑制肝硬化的发展。
        综上所述,银杏叶提取物可改善肝功能,有效抑制肝硬化的发展,其机制可能是通过抗肝细胞凋亡作用来发挥的。
